ntroduction

Proteomic profiling of mitochondria has the potential to provide insights into mitochondrial functions

associated with aging, various metabolic states and diseases such as cancer, diabetes and

cardiovascular disease [1]. Rapid and reproducible isolation of intact mitochondria is crucial for

efficient enrichment and subsequent proteomic analysis of low-abundance mitochondrial proteins [2].

Here we describe a system for the isolation of intact mitochondria from rat PC12 cells using pressure

cycling technology （PCT）.

近年来，线粒体蛋白组学的研究揭示了线粒体相关蛋白与人类的寿命、各种新陈代谢的状态以及很多疾病比如癌症、糖尿病、心血管病等存在一定的关联。对于数量极其有限的线粒体蛋白进行收集并进行蛋白质组学分析，其中至关重要的工作就是从细胞中快速、持续的分离完整的线粒体。本文描述了如何利用压力循环技术（PCT）从鼠PC12细胞中快速分离完整的线粒体。
